The Darkness escapes when Rowena’s spell removes Dean’s Mark of Cain, and its arrival brings a violent outbreak to a Nebraska town. Sam and Dean meet Deputy Jenna while trying to protect survivors, including baby Amara, whose birthmark matches the Mark. Dean discovers a strange bond with the growing Amara: she says he helped free her and that they are bound to help each other. The brothers also face the consequences of their earlier choices as they search for a way to stop her without abandoning people caught in the disaster.

Sam follows visions to Lucifer’s Cage and refuses Lucifer’s offer to become his vessel, even as Lucifer argues he is the only weapon against the Darkness. Castiel eventually accepts Lucifer instead, and the archangel uses his new vessel to confront Amara; she overwhelms him and takes Lucifer out of Cas. The crisis brings Chuck back into the Winchesters’ lives, and he reveals that he is God. Amara is Chuck’s sister, imprisoned by him before creation. When her attacks leave Chuck dying and the sun fading, the brothers work with him, Rowena, Crowley and the reaper Billie on a soul-powered bomb meant to destroy her.

Dean carries the bomb inside himself and approaches Amara ready to die, but he talks to her about the family bond she has lost with Chuck. Amara chooses to heal her brother, restoring the sun and ending the immediate threat. She then gives Dean what he most wants by returning Mary Winchester. Meanwhile, Lady Toni Bevell confronts Sam and Castiel at the bunker: she banishes Cas and fires at Sam, leaving his fate unresolved at the season’s end.
