Peter Parker is a teenager in New York living with Aunt May and Uncle Ben after his scientist parents disappear. At school he is bright but awkward and often bullied; he is also drawn to Gwen Stacy. When Peter finds his father’s briefcase, he learns that Richard Parker worked with Dr. Curt Connors at Oscorp. Searching for his father’s research takes Peter into a restricted laboratory, where a genetically modified spider bites him. The new abilities—strength, agility, speed, and the ability to cling to surfaces—arrive alongside a more personal mystery: why his parents left, and what work Richard was trying to protect.

Connors has lost an arm and is attempting to use cross-species genetics to regenerate limbs. Peter discovers a missing algorithm in his father’s papers and brings it to Connors, revealing his connection to Richard. At home, Peter argues with Ben and leaves; while searching for him, Ben is killed by a thief. Peter uses his powers to track people matching the killer’s description, first acting from grief rather than a settled sense of responsibility. He creates a mask and suit, then builds mechanical web-shooters using an Oscorp formula. His new life intersects with Gwen’s family when dinner reveals that her father, police captain George Stacy, distrusts Spider-Man. Peter tells Gwen his identity and they begin a relationship.

At Oscorp, Connors’ superior Rajit Ratha demands human trials of the limb-regeneration formula. Connors refuses to rush the testing, fearing harm to people, but after Ratha fires him he injects the serum into himself. His missing arm grows back; the transformation also turns him into a reptilian creature with violent impulses. When Connors attacks Ratha on the Williamsburg Bridge, Peter intervenes and saves people, but the city’s response treats both Spider-Man and the Lizard as threats. Peter learns that Connors is planning to release a chemical agent from Oscorp Tower, one that would turn the city’s population into reptilian hybrids. Captain Stacy discovers Peter’s identity when police corner him during their pursuit.

Stacy lets Peter pursue Connors, while Gwen works on an antidote. Connors prepares to spread his formula from Oscorp Tower, and Peter has to reach the apparatus before that plan turns an entire city into unwilling test subjects. Stacy joins the fight, giving Peter an opening to replace the planned payload with the antidote. The cure reverses the transformations, including Connors' own. Mortally wounded, Stacy asks Peter to keep away from Gwen to protect her. Peter attempts to honor the request, and Gwen is hurt by his withdrawal. Their last exchanges suggest that the promise will not hold, leaving the personal consequences unsettled even after the immediate threat has ended.

The film’s New York is built around overlapping systems: school, police, family, and Oscorp research all shape Peter’s choices. His father’s work helps Connors but also gives Peter a route into a laboratory where a single bite changes his body. The web-shooters make his heroism partly a product of engineering and improvisation, not only an inherited power. Gwen is not simply outside the action; her scientific work produces the antidote the city needs. Connors’ transformation begins with a real wish to repair an injury and becomes dangerous when he treats a whole population as test subjects, echoing the same institutional pressure that first led him to rush.

Uncle Ben's death follows an argument that Peter cannot take back, making the early hunt for a thief an attempt to find an outward target for guilt. Aunt May still has a household to hold together while Peter disappears, returns injured, and struggles to explain himself. Those ordinary pressures give his secret identity a cost beyond avoiding arrest. The people caring for him do not automatically know what has happened or why he behaves differently. Being able to climb a building does nothing to make a difficult conversation easier. Peter has to learn what his abilities mean while also facing the responsibilities he already had before the bite.

The bridge rescue brings Peter's abilities into contact with a frightened child rather than another superpowered figure. Later, help travels in the opposite direction: the child's father organizes crane operators to give the injured Spider-Man a route toward Oscorp Tower. The city is therefore more than a collection of people waiting to be saved. Strangers can notice what he has done and decide to act themselves. That response also complicates the police view of an irresponsible masked intruder. Peter's methods remain improvised, but the rescue has created a connection that becomes useful when his own strength is no longer enough to carry him through the crisis.

Andrew Garfield's Peter is restless and often defensive, while Emma Stone's Gwen can meet him in the scientific part of his life as well as the romantic one. Their shared access to Oscorp makes their involvement in the crisis credible within the story: she can understand the laboratory work, and he has unknowingly supplied an important piece of it. Rhys Ifans gives Connors an injury and a professional frustration before the creature emerges. The hope behind the experiment remains recognizable even when its outcome becomes dangerous. Peter's parents are still an unanswered part of this film, so stopping Connors closes one disaster without turning the family's disappearance into a solved case.
